چکیده

For many years, sodium caprylate has been used as a suitable stabilizer to retain biological activity of pharmaceutical proteins such as human albumin solutions during pasteurization. A simple method of turbidity assay was applied for evaluation of sodium caprylate as a stabilizer to protect biological activity of infusible platelet membrane product during pasteurization at 60°C for 20 h. Concentration of sodium caprylate of samples were 0.4, 0.2, 0.1 and 0.05 M. After pasteurization, turbidity of samples was measured with spectrophotometer at optical density of 450 nm. The results were found 0.662, 1.890, 2.300, 2.365 respectively and it was concluded that sodium caprylate concentration of 0.4 M is suitable for pasteurization of infusible platelet membrane product and also turbidity assay is simple and efficient method for evaluation of this process.

بررسی اثر سمیت زایی غشاء پلاکت قابل تزریق(IPM ) در موش: یک گزارش کوتاه

Background and Objective: Blood platelet units are generally stored in blood banks for 3–5 days, afterwards they are discarded. Prepared infusible platelet membrane (IPM) from fresh or outdated human platelets correct the prolonged bleeding times in thrombocytopenic animals such as rabbits. Abnormal Toxicity is an European Pharmacopoeia standard for assessment of biological products and th...

Infusible Platelet Membrane versus Conventional Platelet Concentrate: Benefits and Disadvantages

Blood transfusion centers are under considerable pressure to produce platelet concentrates with a shelf life limit of 3 to 5 days. Many approaches have been investigated experimentally to produce new hemostatically active platelet products that are capable of long term storage. In this article infusible platelet membrane will be explained as a platelet substitute versus conventional liquid-stor...

Study of the Heat-Treated Human Albumin Stabilization by Caprylate and Acetyltryptophanate

Sodium salt of N-acetyl-DL-tryptophan (AT) and octanoic acid (caprylic acid CA) individually and in combination were studied as thermal stabilizer with different concentrations in heat treatment of human albumin solution. HPLC protein profiles before pasteurization of albumin showed less than 1% polymer. In free stabilizer albumin solution during pasteurization visible clot was formed. But by u...

Flow Cytometric Measurement of CD41/CD61, CD42b Platelet Receptors and Platelet Factor 3 Activity in Lyophilized Infusible Platelet Membrane Preparation

Background: The short life time of human platelet units has led to a chronic shortage of fresh platelets in blood transfusion centers. Many approaches have been investigated experimentally to produce new hemostatically active platelet products that are capable of long term storage. Infusible platelet membrane (IPM) prepared from fresh or outdated human platelets have been developed as an altern...

The surface markers and survival rate of platelets during storage at 4°C: The influence of sodium octanoate

Background: Storage of platelet concentrates (PCs) at room temperature (20-24°C) limits its storage time to 5 days due to the destructive effects of platelet storage lesion (PSL) and bacterial contamination. Although prolonged storage of platelets (PLTs) at 4°C reduces the likelihood of bacterial contamination and PSL levels, it is accompanied by an increase in the clearance rate and changes in...
